The Ministry of Transport has responsibility for air, land and sea transport.
Flight MH370 was an aviation disaster and therefore involved the Department of Civil Aviation (Malaysia) which is "an agency under the Ministry of Transport (MOT) Malaysia to provide safe, efficient and orderly flow of air transportation, and to regulate aviation activities in Malaysia." The DCA is now CAAM.
When Malaysia Airlines flight MH370 went missing on Saturday, 8 March 2014, the Minister of Defence, Hishammuddin Hussein was also Acting Minister of Transport.
Hishammuddin Hussein was initially responsible for the investigation into the loss of MH370 and, as spokesperson for the Department of Transport, delivered many of the Media Statements in 2014.
That role has been passed to his successors (see below).

As noted above, Hishammuddin Hussein was Acting Minister of Transport when flight MH370 went missing.
